Aspergillus fumigatus strain NEIST P13 BioVector NTCC质粒载体菌种细胞基因保藏中心 A fungal strain was selected from the microbial repository which could produce a high yield of L-amino acid oxidase. 18SrRNA, ITS1, 5.8SrRNA ITS2, and partial 28 S rRNA sequencing and phenotypic characteristics indicate that it belong to the species Aspergillus fumigatus (designated as P13). Maximum production of enzyme (59.55 x 10-3 U/mg dry cell mass) was obtained in a medium containing 10 g/L glucose, 4 g/L yeast extract, and 4 g/L ammonium sulfate, with 20 mmol/L of L-threonine as the inducer. The optimum temperature for enzyme production was 30 degrees C at pH 7.0, with a shaking speed of 200 r/min. At 96 h, the enzyme activity was maximum. The A. fumigatus P13 L-amino acid oxidase accepts a broad substrate range, and the maximum enzyme activity (20.41 x 10-3 U/mg dry cell mass) was obtained with 50 mmol/L of L-tyrosine. In the literature, no reports have been found regarding the production of L-amino acid oxidase by A. fumigatus. The enzyme showed enantiomerically pure amino acid formation, which has tremendous demand in industrial applications.

Medium
POTATO DEXTROSE AGAR
Infusion from potatoes (see below) 1000.0 ml
Glucose 20.0 g
Agar 15.0 g
Potato infusion:
Boil 200 g scrubbed and sliced potatoes in 1000 ml water for 1 hour. Pass through fine
sieve. Avoid new potatoes.

Growth Conditions
Temperature: 30°C
Atmosphere: Aerobic
Propagation Procedure
1. Open vial according to enclosed instructions.
2. Using a single tube of #3 broth (5 to 6 mL), withdraw approximately 0.5 to 1.0 mL with a Pasteur or
1.0 mL pipette. Rehydrate the entire pellet.
3. Aseptically transfer this aliquot back into the broth tube. Mix well.
4. Use several drops of the suspension to inoculate a #3 agar slant and/or plate.
5. Incubate the tubes and plate at 30°C for 24 hours.
