NanoLuc Genetic Reporter Vectors


NanoLuc®NlucluciferaseNanoLuc® 萤光素酶)是一个经过基因工程改造的小分子酶(19.1kDa),是性能卓越的生物发光报告基因。该酶比萤火虫(Photinus pyralis)或海肾(Renilla reniformis)萤光素酶的发光亮 100 倍,它使用一种新型底物——furimazine,可产生高强度、辉光型发光。生物发光反应不依赖 ATP,同时抑制背景发光以获得最高检测灵敏度。

目前已构建多种类型的 NanoLuc® 萤光素酶报告基因载体,以满足不同的实验需求。非融合的 Nluc 提供了最大的光输出和灵敏度,NanoLuc®-PESTNlucP)将蛋白表达水平变化与转录活性改变密切偶联,并增加信噪比,融合 N- 末端分泌信号(secNluc)的 NanoLuc® 萤光素酶适用于分泌型报告基因试验。使用 Nano-Glo® 萤光素酶检测试剂,发光量在跨越 1,000,000 倍的酶浓度范围内呈线性关系,且信号半衰期≥ 2 小时。

NanoLuc® 萤光素酶具有许多使之成为出色报告基因蛋白的物理特性:
分子量非常小,单体酶(171 个氨基酸;513bp
热稳定性高(Tm=60
在很宽的pH 范围内有活性(pH 6-8
没有翻译后修饰或二硫键
均匀分布在细胞中
发射光谱非常适合于生物发光共振能量转移(BRETλmax=465nM)。

为满足利用报告基因研究转录调控,我们提供多种表达不同类型 NanoLuc® 萤光素酶(非融合型NlucPEST 不稳定型NlucP、分泌型secNluc)的质粒载体。不同的 pNL 载体用途如下:
• pNL1
:克隆已知或预测的启动子区域
• pNL2
:克隆已知或预测的启动子区域,并通过潮霉素(Hygromycin)筛选建立一个稳定克隆的细胞系
• pNL3
:克隆不需要基本启动子的结合位点或应答原件(如 pNL3.2.NF-κB-RE 载体)
提供非融合和分泌型 Nluc 对照质粒。pNL 载体系列以pGL4 为基础设计,以方便从现有的质粒上进行序列转移。载体骨架设计还去掉了许多转录因子结合位点和其他潜在的调控元件以减少反常结果的发生。对Nluc基因突变体进行了密码子优化,并去除了许多潜在调控元件或其他不必要的功能(如常见的限制性内切酶位点)。
