The PSAM2-PAX7 vector contained a tet-responsive
element to induce Pax7 expression by doxycycline (Dox)
and an internal ribosome entry site (IRES)-EGFP reporter to assess
transgene expression through green fluorescence. Inducible
expression of Pax7 was confirmed based on EGFP (Pax7) expression,
following an overnight incubation of ES/iPS cells with 0.75 μg/
mL dox. Cells were then harvested and differentiated as embryoid
bodies (EBs).
