The Expression Vector pVB4
The pVB4 vector is developed for high level protein production in Escherichia coli and other Gram negative
bacteria.
Expression
The pVB4 vector features the Pm/xylS expression cassette for controlled high-level recombinant gene expression.
Induction
For maximum expression induce with 2 mM m-toluic acid.
Cloning advice
A multiple cloning site with different restriction sites for easy cloning of gene of interest is positioned
downstream of the Pm promoter. To achieve optimal expression the gene should ideally be cloned in with the use
of AflIII. This ensures that the start of the gene will be in optimal distance from the ribosome binding site (RBS).
Replication and host range
The vector contains oriV and wild type trfA from RK2 which gives the vector broad-host-range for high-level
protein production in Escherichia coli or other Gram negative bacteria. The copy number of this vector in E. coli is
5-7 copies per chromosome.
The vector can be established in its hosts by electroporation, transformation or by taking advantage of oriT for
conjugal transfer.
pVB4 contains the tetA and tetR genes that confer tetracyclin resistance.
