Introduction The 293-EBNA cell line stably expresses the Epstein Barr Virus (EBV) EBNA-1 gene
from pCMV/EBNA. Expression of the EBNA-1 gene is controlled by the CMV promoter
and is high-level and constitutive. For more information about the pCMV/EBNA plasmid,
please refer to our World Wide Web site (www.invitrogen.com) or call Technical Service
(see page 8).
The 293-EBNA cell line is intended for use with vectors containing an EBV origin of
replication (oriP)(Young et al., 1988). Vectors containing an oriP region can be
maintained episomally in 293-EBNA cells.
Parental Cell Line The table below provides a brief description of the source of the parental cell line used
to generate the 293-EBNA cell line. The parental cell line was obtained from the
American Type Culture Collection (ATCC). The ATCC number for the cell line is
included. For further information about the parental cell line, please refer to the ATCC
Web site (www.atcc.org).

293-EBNA cells stably express the neomycin resistance gene from pRSV4neo (Lambert
et al., 1988) and should be maintained in medium containing Geneticinô (G418 sulfate)
at the concentration listed below. Expression of the neomycin resistance gene in 293-
EBNA cells is controlled by the Rous Sarcoma Virus long terminal repeat (RSV LTR).
For more information about pRSV4neo, please refer to Lambert et al., 1988.

The table below provides the recommended complete medium, freezing medium, and
antibiotic concentration required to maintain and culture 293-EBNA cells.

Important
Guidelines
ï FBS does not need to be heat inactivated for use with these cell lines.
ï Cell lines should be maintained in medium containing G418 at the concentration listed
above.
ï If cells are split at a 1:5 to 1:10 dilution, they will generally reach 80-90% confluence
in 3-4 days.
