These mice harbor the Transgenic Adenocarcinoma of the Mouse Prostate (TRAMP); PB-Tag Line 8247 transgene. Mice hemizygous for the TRAMP transgene develop progressive forms of prostate cancer with distant site metastasis and exhibit various forms of disease from mild intraepithelial hyperplasia to large multinodular malignant neoplasia.

In 2019, sequencing performed by The BioVector NTCC Inc. discovered that greater than 20 copies of the Tg(TRAMP)8247Ng transgene (hemizygous) integrated on chromosome 16 (~18,993,000-19,272,880; mouse mm10). The region surrounding the integration site has duplicated. Currently, there are no genes annotated in this region.

Detailed Description
TRAMP hemizygotes can exhibit prostatic epithelial neoplasia (PIN) by 12 weeks of age. Tumors, appearing as well differentiated adenocarcinoma, can arise by 24 weeks of age, mostly in the dorsal and lateral lobes of the prostate. By 30 weeks of age, most hemizygous mice will display evidence of metastatic spread to the lymph nodes and/or lungs, phylloides appearance of some tumors, and seminal vesicle invasion. Tumors exhibit elevated levels of nuclear TRP53 and decreased androgen receptor expression. Note that pure C57BL/6 TRAMP hemizygotes may live to 52 weeks of age or longer.

Development
The TRAMP (Transgenic Adenocarcinoma of Mouse Prostate) transgenic construct was designed with simian virus 40 (SV40) early genes (large and small tumor antigens, Tag) under the control of the rat probasin promoter. The founder mouse (line 8247) described in the primary publication is one that developed a large prostate tumor by 10 weeks of age and was generated on a C57BL/6 background. The donating investigator maintains the strain by breeding hemizygous females with C57BL/6 males. The strain has been backcrossed to C57BL/6 for 20+ generations.

TRAMP stands for TRansgenic Adenocarcinoma Mouse Prostate. Lines 5666 and 8079 were also produced.
Transgenic mice on a C57BL6 background develop progressive forms of prostate cancer with distant site metastasis. Forms of disease range from mild intraepithelial hyperplasia to large multinodular malignant neoplasia. Tumors are detected in the prostate as early as 10 weeks of age. The tumors have elevated levels of nuclear TRP53 and decreased androgen receptor expression.

Breeding Considerations
When maintaining our live colony, C57BL/6J inbred mice are bred to hemizygous mice. Most commonly, but not exclusively, this is C57BL/6J females bred to hemizygous males. Expected coat color from breeding is black. [2020]
