Red/ET recombination relies on homologous recombination in vivo in E.coli andallows a wide range of modifications on DNA molecules of any size and at anychosen position. Homologous recombination is the exchange of genetic materialbetween two DNA molecules in a precise, specific and accurate manner.Homologous recombination occurs through homology regions, which are stretches ofDNA shared by the two molecules that recombine. Because the sequence of thehomology regions can be chosen freely, any position on a target molecule can bespecifically altered. Red/ET recombination allows you to choose homology arms asshort as 50 bp for homologous recombination, which can easily be added to afunctional cassette by long PCR primers.Full-length Rac prophage protein RecE and its partner RecT mediate highly efficientlinear-to-linear homologous recombination mechanistically distinct from conventionalrecombineering mediated by Redαβ from lambda phage (Fu et al. 2012).E. coli strain BioVector® GB05-dir harbours an arabinose inducible ETgA operon (full lengthrecE, recT, redγ, and recA) at the ybcC locus. The strain was successfully used todirectly clone gene clusters larger than 10 kb of secondary-metabolite pathways fromPhotorhabdus luminescens, a gram negative rod-shaped bacterium, into expressionvectors (Fu et al. 2012).1.
