This vector requires cir+ hosts. The centromere fragment (KlCEN2) comes from chromosome II. The plasmid is otherwise identical to KEp6. Priming sequence for one of the TetR cloning site is also present at the side of KlCEN2. You can't use it for confirming the insert.
