Phosphatidylserine (PS) Detection: The primary function of the plasmid is to localize and track PS, an important anionic phospholipid that is typically found on the inner (cytosolic) leaflet of the plasma membrane in healthy cells. Its exposure on the outer leaflet often serves as a signal for apoptosis (programmed cell death) and other biological processes.
GFP Fusion Protein: The expressed GFP-Lact-C2 fusion protein binds with high affinity and specificity to PS in a calcium-independent manner. When expressed in cells, the GFP tag allows researchers to visualize the location of PS within the cell using fluorescence microscopy.
Localization: The probe primarily labels the plasma membrane, but it can also decorate internal organelles like endosomes, reflecting the presence of PS in these various cellular compartments.
Specificity: The C2 domain specifically interacts with PS and is selective for PS in different membrane environments, making it a reliable tool for distinguishing PS from other phospholipids.
