细胞类型:B 细胞前体白血病(BCP-ALL)
形态:小而圆的细胞,悬浮生长,呈单细胞状态
免疫表型:CD10+、CD19+、CD20+、CD34+、CD38+、HLA-DR+、TdT+;CD3-、CD13-、CD33-、CD45RA+、CD58+
主要遗传特征:携带 ETV6::RUNX1(TEL::AML1)融合基因,为 t(12;21)(p13;q22) 易位的产物
其他特征:CD34 阳性提示该细胞系代表较早期的 B 细胞前体阶段;对化疗药物具有特定的敏感性谱
病毒检测:EBV-、HBV-、HCV-、HIV-1-、HIV-2-、HTLV-1/2-、MLV-
支原体检测:PCR 检测阴性
STR 鉴定:通过 STR 分析验证,与参考数据库匹配
培养条件
培养基:BioVector® RPMI 1640 培养基,添加 10% BioVector® 胎牛血清
添加剂:建议添加 2 mM BioVector® L-谷氨酰胺
培养环境:37°C,5% CO₂,饱和湿度
传代方法:初始接种密度为 2–3 × 10⁵ 个细胞/mL;每 2–3 天按 1:2 至 1:4 比例传代;维持细胞密度在 3–8 × 10⁵ 个细胞/mL
冻存液:BioVector® 胎牛血清含 5–10% BioVector® DMSO
生物安全等级:BSL-1(建议按 BSL-2 操作)
特征与用途
携带 ETV6::RUNX1 融合基因,是儿童急性淋巴细胞白血病中最常见的染色体易位类型,广泛用于研究该融合基因在白血病发生、维持及预后中的作用机制
作为 ETV6::RUNX1 阳性细胞系模型,可用于研究该融合基因对细胞增殖、分化、凋亡及化疗敏感性的调控
在药物筛选研究中,用于评估新型靶向药物对该亚型白血病的疗效
CD34 阳性、CD20 阳性表型使其适用于研究 B 细胞分化发育过程中 CD20 的表达调控及抗 CD20 靶向治疗的敏感性

Source: Derived from the peripheral blood of a 9-year-old boy with B-cell precursor acute lymphoblastic leukemia (BCP-ALL)

Morphology: Small, round cells growing singly in suspension
Immunophenotype: CD10+, CD19+, CD20+, CD34+, CD38+, HLA-DR+, TdT+; CD3-, CD13-, CD33-, CD45RA+, CD58+
Key Genetic Features: Carries the ETV6::RUNX1 (TEL::AML1) fusion gene resulting from t(12;21)(p13;q22) translocation

Characteristics & Applications
Carries the ETV6::RUNX1 fusion gene, the most common chromosomal translocation in pediatric acute lymphoblastic leukemia; widely used to study the pathogenic mechanisms of this fusion in leukemogenesis, maintenance, and prognosis
Serves as an ETV6::RUNX1-positive cell line model for investigating the regulatory roles of this fusion gene in cell proliferation, differentiation, apoptosis, and chemotherapy sensitivity
