一、 背景与来源说明 / Background & Origin

BioVector® MUTZ-8 是一种建立自一名63岁患有继发性急性粒-单核细胞白血病(Acute Myelomonocytic Leukemia, AML FAB-M4)女性患者外周血的人源白血病细胞株。该患者在出现骨髓增生异常综合征(MDS)25年后演变为急性白血病。

MUTZ-8 细胞株具有显著的遗传学与生物学特征:

  1. 细胞因子严格依赖性:高度依赖 GM-CSF 或相关细胞因子(如 IL-3、SCF)维持体外生存与增殖。

  2. 关键基因突变与染色体变异携带经典的 JAK2 V617F 纯合突变 以及 TP53 突变,并伴有 5q31 缺失$\text{t}(5;11)(\text{q21};\text{q10})$ 非相互易位等复杂染色体核型变异。

三、 培养条件与传代指南 / Culture & Handling Protocols

1. 推荐培养基 / Complete Growth Medium

  • 基础培养基BioVector® $\alpha$-MEM Medium(含核苷与脱氧核苷) 或 BioVector® RPMI-1640 Medium

  • 血清浓度:20% 灭活 BioVector® Fetal Bovine Serum (FBS) + 1% 双抗(Penicillin-Streptomycin)

  • 必需细胞因子 / Vital Supplement必须添加重组人粒细胞-巨噬细胞集落刺激因子 rhGM-CSF (10 ng/mL)(或 10% 5637 细胞条件培养基)。缺乏细胞因子或接种密度过低会导致快速凋亡或诱导因子不依赖性突变株的生长。

  • 培养环境:37 摄氏度,5% $\text{CO}_2$,高湿度培养箱

2. 复苏与传代操作 / Thawing & Subculture

  1. 复苏:将冻存管快速置于 37 摄氏度水浴中融化(1–2 分钟)。加入 8–10 mL 预热并补充有 rhGM-CSF 的 BioVector® Complete Medium,120 x g 离心 5 分钟,弃上清后重悬接种。初复苏时建议接种密度稍高(约 $0.8 \times 10^6 \text{ cells/mL}$)以协助恢复。

  2. 传代与密度控制

    • 维持密度:建议维持细胞密度在 $5 \times 10^5 \text{ cells/mL}$$1.0 \times 10^6 \text{ cells/mL}$ 之间(最高密度约 $1.2 \times 10^6 \text{ cells/mL}$)。

    • 换液传代:每周按 1:2 的比例分瓶或补液传代 1–2 次。无需 Trypsin 消化。倍增时间约为 3–5 天。

Thawing and Subculture Protocol

  1. Thawing: Thaw rapidly in a 37 degrees Celsius water bath. Resuspend in pre-warmed BioVector® Complete Medium supplemented with rhGM-CSF and centrifuge at 120 x g for 5 minutes. Initial seeding density after thaw should be $\sim 0.8 \times 10^6 \text{ cells/mL}$.

  2. Passaging:

    • Maintain cell density between $0.5 \times 10^6 \text{ cells/mL}$ and $1.0 \times 10^6 \text{ cells/mL}$.

    • Subculture 1:2 once or twice weekly by dilution or centrifugation. Doubling time is approximately 3–5 days.
