Description: The pKK233-2 Expression Vector Kit is designed for
direct cloning of eukaryotic genes in E. coli: The plasmid contains
the highly expressed trc promoter (17 base pair spacing between the
trp -35 region and the lac UV5-10 region), the lacZ ribosome
binding site and ATG initiation codon (2). To prevent unstable
replication, the strong rrnB transcription terminator has been
introduced downstream of the Multiple Cloning Site. Digestion with
Nco I exposes the start codon for direct ligation and expression of
foreign proteins. Eukaryotic gene fragments lacking a prokaryotic
ribosome binding site and/or an ATG can be inserted in the correct
reading frame by using the three different Nco I linkers included
in the kit. The Nco I recognition sequence, CCATGG, commonly occurs
at the initiation codon of eukaryotic genes, allowing direct
ligation to the vector (2,3). The pKK233-2 vedor has been
successfully used to express truncated forms of the cI repressor
protein from lambda (2). Herpes Simplex Type 1 glycoprotein D (2),
rat lipocortin (4) and chicken triose phosphate isomerase (5).
