■ 产品概述：

Stbl2 Competent Cells have been prepared by apatented modification of the procedure of Hanahan . These cells are suitablefor the cloning of unstable inserts such as retroviral sequences or directrepeats .

For optimal performance, perform expression in S.O.C.Medium and incubation on antibiotic plates at 30°C. Stbl2 Competent Cells are aderivative of JM109. The mcrA mutation and the mcrBC-hsdRMS-mrrdeletion allowfor the cloning of genomic sequences that are methylated. These cells are notcapable of blue/white selection with plasmids containing α-complementationsequences.

■ 产品储存：

-80°C 保存，避免反复冻融。

■ 使用步骤：

1. 把感受态细胞置于冰中融化。

2. 把100uL的感受态细胞移至灭菌处理的试管内。

3. 加入用于转化的DNA（10 ng以下）。

4. 冰中放置30分钟。

5. 42℃放置45秒。

6. 冰中放置2～3分钟。

7. 加入30℃预温好的SOC培养基，使终体积为1 ml。

8. 30℃振荡培养1小时（160～225 rpm）。

9. 取适量涂布琼脂平板培养基。

10. 30℃过夜培养。

■ 注意事项：

1. 请用低温运输。

2. 不立即使用的感受态细胞请在-80℃下保存（融化后的感受态细胞不能再冻结保存）。

3. 转化时请使用传热性能较好的试管。一般的 1.5 ml 微型离心管也可使用，但转化效率会有所下降（此时请在使用方法5.的42℃下放置60秒）。

4. 对100 uL的感受态细胞，用于转化的质粒DNA量请控制在10 ng以下，并保证DNA 溶液的体积在20 uL以下。否则会影响转化效率。

5. 使用SOC培养基的地方也可使用 LB 培养基，但此时转化效率会有所下降。
