pEC1 Staphylococcus gene replacement plasmid. Restriction maps of the ermB cassette fragments. Plasmids pEC1-pEC4 contain the 1.45 kb ClaI ermB fragment of Tn551 cloned into the AccI restriction site of pUC18 (pEC1, pEC2), or into the ClaI restriction site of pBluescript KS+ (pEC3, pEC4). Plasmids pEC5 is composed of the 2.23 HpaI–SalI ermB fragment cloned into EcoRV–SalI cleaved pBluescript KS+. Plasmids pEC6 and pEC7 consist of the 2.31 kb HpaI–BglII ermB fragment cloned into pUC19 cut with BamHI–HincII (pEC6) or BamHI–SmaI (pEC7).
